Akiba's Trip: Undead and Undressed has multiple gameplay routes.
There are four total routes, one each for Tohko, Shizuku, Rin, and Shion.
The game is split into three chapters. The first two chapters are always the same, with only slight deviations possible. Each girl has their own unique chapter 3 route.
The dialogue choices you make in chapters 1 and 2 determine which route you'll take in chapter 3.
You'll need 50 or more points of affection with a girl in chapters 1 and 2 to reach her chapter 3 route. If you don't get at least 50 points of affection with any girl by the end of chapter 2, you'll be placed on Tohko's route.
A girl's affection level is raised by picking specific dialogue choices during cut scenes.
Picking a dialogue choice that raises a girl's affection does not lower any other girl's affection.
Affection is a hidden value you'll have to keep track of on your own. However, it's pretty easy to do so, especially with the information in this guide.
Dialogue choices that raise a girl's affection will do so by either +5, +10, or +15. There is not an equal number of choices per girl. Some girl's routes are harder to reach than others.
Each girl's route has two endings: a "normal ending" (which can be considered that girl's bad ending) and a "true ending" (which can be considered that girl's good ending). Don't get confused by the phrase "true ending", as there isn't one canon true ending, it's just the label the game uses for a girl's good ending.
You'll need 100 or more points of affection with a girl by the end of chapter 3 to reach her true ending. If you get less than 100 points of affection by the end of chapter 3, you'll receive the normal ending of whichever girl's route you're on.
The following table shows how many dialogue choices are available to raise each girl's affection in each chapter. It also shows the total number of affection points available for each girl in each chapter.

If a tie-breaker needs to be instituted to decide which route you'll be put on in chapter 3, the order of priority is as follows:
Tohko > Shizuku > Rin > Shion
Kati doesn't have a route or ending in the original release of the game.
During the first playthrough of the game, there's no visual indicator about how dialogue choices affect a girl's affection. However, future playthroughs will display an icon of one of the girls' faces next to dialogue choices that boost that girl's affection.
Nana has a special ending that can be unlocked by successfully completing a series of specific side missions known as Sister Missions.
Unlike normal side missions, you accept Sister Missions by speaking to Nana (or Pops, at one point) at specific points during the game. You'll need to have completed all prior Sister Missions to be given the next one. Nana's overhead name tag will be red if she has a mission to give you.
Nana's ending does not change the ending of the route you're on, it's just an extra scene that plays after the route ending you earned.
